RT Dissertation/Thesis T1 Selective recognition of threeway DNA junctions with designed peptidomimetics A1 Alcalde Ordóñez, Ana K1 DNA structures K1 drugs K1 peptide ligands K1 stereoselective synthesis of dinuclear Cu(II) AB Non-canonical DNA structures, such as three-way DNAjunctions (3WJs), formed during DNA replication, have emergedas promising alternative DNA targets for the development ofnew drugs with attractive therapeutic properties. In this context, supramolecular helicates have shown high selectivity andbinding affinity for 3WJs, but their use in this field is hamperedby the lack of synthetic versatility and a general strategy to theirenantioselective synthesis. In this thesis, we describe thestereoselective self-assembly of helicates derived from peptideligands equipped with 2,2’-bipyridine coordinating residues, aswell as the implementation of a new strategy that relies on thecooperative 3WJ-binding mechanism of natural proteins, to yieldnew systems with selective 3WJs recognition properties.
