AB Alzheimer disease (AD) is a devastating illness that affects populations worldwide, and for which an effective cure has yet to be found. We have studied in depth the effect of various factors like pH and ligand structure on the inhibition of BACE-1, one of the enzymes that produce the Aβ peptides, whose aggregation is a hallmark of this disease. The results of this study helped us to obtain drug leads that inhibit not only the aforementioned enzyme but some other AD therapeutic targets at the core of this illness. This study successfully identified several compounds with true multitarget potential against AD at the amyloidogenic and cholinergic levels.La enfermedad de Alzheimer (EA) es una enfermedad de alcance mundial para la que todavía no se ha encontrado un tratamiento efectivo. Hemos estudiado el efecto de varios factores como el pH y la estructura de los ligandos en la inhibición de BACE-1, uno de los enzimas que interviene en la formación de péptidos Aβ, cuya agregación es clave en el desarrollo de dicha enfermedad. Los resultados de estos estudios nos han ayudado a obtener compuestos cabeza de serie capaces no sólo de inhibir la actividad de BACE-1, sino también de inhibir otras dianas clave de la ruta patológica de la EA. Hemos identificado varios compuestos con potencial real como fármacos multidiana en el tratamiento de la EA a niveles amiloidogénicos y colinérgicos.
