RT Dissertation/Thesis T1 Novel approaches for the discovery of pharmacogenetic biomarkers of chemotoxicity in patients with colorectal cancer A1 Simões, Ana Rita Santos K1 colorectal cancer K1 adverse drug reactions K1 pharmacogenetics K1 pharmacogenomics K1 personalised medicine AB Colorectal cancer (CRC) is often linked to adverse drugreactions (ADRs), which can be severe or even fatal. To addressthis issue, pharmacogenetic studies have emerged to helppersonalise treatments based on molecular information. However, these have been limited by a lack of standardizeddata, the complexity of ADR phenotyping and the need fornovel strategies and functional assays. Therefore, the aim ofthis project was to find and validate new CRC chemotoxicitybiomarkers, using state-of-the-art approaches. We haveconducted different omic analyses to identify low to highpenetrancevariants and performed in vitro assays on a variantof interest. Our work successfully identified severalcandidate toxicity variants/genes that might be influencing thedevelopment of ADRs in CRC patients.
